چکیده

Abstract Background and Aims Patients with end stage renal disease (ESKD) on hemodialysis (HD) have a high risk of cardiovascular (CV) events, this being mostly related to the presence uraemic cardiomyopathy. A deregulation in expression various microRNAs (miRNAs: non-coding RNAs that regulate post-transcriptional gene expression) has previously been implied pathological cardiac remodelling. Nevertheless, scarce evidence accrued so far possible significance CV morbidity mortality HD patients, relationship uremic Method We run pilot, prospective, multicentre cohort study involving 74 ESKD patients undergoing chronic from 3 different hospitals Italy Greece. underwent thorough clinical, laboratory echocardiography assessment were then prospectively followed for 24 months or until occurrence composite endpoint (CV all-cause) non-fatal events. Through systematic review literature, we identified small panel miRNAs (30a-5p, 23a-3p,451a let7d-5p), which levels are known be altered either major disorders kidney failure. measured blood group matched healthy controls. Results 23a-3p (p<0.0001), 451a (p = 0.001), 30a-5p 0.003) let7d-5p <0.0001) all reduced as compared Significant correlations found between indexes dysfunction such Vmax, TAPSE E/E’, well some other parameters uric acid, sodium, potassium, HDL CRP. During follow-up, 30/74 (40.5%) reached endpoint. In these individuals, but significantly at baseline (p<0.0001). As showed by multivariate Cox-regression analyses, 23a-3p, 451a, predictors (HR ranging 0.943 0.995; p 0.05 0.02) Kaplan-Meier analyses confirmed faster progression subjects below an optimal ROC-derived cut-off value 0.001 <0.0001; crude HRs 7.95 8.61; Figure 1). Conclusion Our demonstrated evaluation very circulating (30-5p, 23a–3p 451a) may impart important prognostic information respect risk. Although biological meaning their remains unanswered, preliminary findings set larger investigation generalize usefulness biomarkers, therapeutic targets.

Serum Gamma-Glutamyltransferase Levels Predict Clinical Outcomes in Hemodialysis Patients

BACKGROUND Gamma-glutamyltransferase (GGT) is a biomarker of liver injury. GGT has also been reported to be a marker of oxidative stress and a predictor of mortality in the general population. Hemodialysis (HD) patients suffer from oxidative stress. The aim of our study was to investigate the relationship between serum GGT levels and clinical outcomes in HD patients. METHODS A total of 1,634 ...

Association Between Circulating Ketone Bodies and Worse Outcomes in Hemodialysis Patients

BACKGROUND Cardiovascular disease is the leading cause of morbidity and mortality in patients receiving hemodialysis. Systemic metabolic perturbation is one of the hallmark abnormalities in patients at high cardiovascular risk. We sought to determine the relationship between circulating ketone body and clinical outcomes in patients with prevalent hemodialysis. METHODS AND RESULTS We retrospec...

Cardio-miRNAs and onco-miRNAs: circulating miRNA-based diagnostics for non-cancerous and cancerous diseases

Cardiovascular diseases and cancers are the leading causes of morbidity and mortality in the world. MicroRNAs (miRNAs) are short non-coding RNAs that primarily repress target mRNAs. Here, miR-24, miR-125b, miR-195, and miR-214 were selected as representative cardio-miRs that are upregulated in human heart failure. To bridge the gap between miRNA studies in cardiology and oncology, the targets a...

Early-phase circulating miRNAs predict tumor recurrence and survival of hepatocellular carcinoma patients after liver transplantation

Post-liver transplantation tumor recurrence is a major challenge for hepatocellular carcinoma (HCC) recipients. We aimed to identify early-phase circulating microRNAs after liver transplantation for predicting tumor recurrence and survival of HCC recipients. Circulating microRNA profiles at early-phase (2-hour after portal vein reperfusion) after liver transplantation were compared between HCC ...

Low 17β-estradiol Levels Are Better Inducers of Regulatory Conditioned T Cells In-Vitro

Background: 17β-estradiol (E2) has been known to modulate immune response. Recent studies indicate that E2 at pregnancy level plays a role in regulating T cell response. Objective: To investigate the optimum dose of E2 (from 10-9 to 10-7 M) in mediating the generation of regulatory T cells (Tregs), using naïve human CD4+ T cells from healthy women. Methods: Naïve peripheral T cells were purifie...
